Design Notes

We added a short sequence CCTGCAGG before bjGFP to ensure proper distance between rbs and bjGFP, leading to higher translation efficiency.


Source

The promoter PgsiB comes from Bacillus subtilis subsp. subtilis str. 168.

The rbs sequence information is from BBa_K2963006 and the sequence itself is synthetic.

The bjGFP is originally chemical synthesized and we amplified this sequence from the plasmid pRJPaph-bjGFP.

The rrnB terminators sequence information is from BBa_K3568009 and the sequences for rrnB T1 and T2 are carried by the plasmid pXMJ19.
